This is the most important thing to understand about GABA supplements, and most supplement companies will not tell you about it.
Your brain is protected by the blood-brain barrier (BBB), a selective membrane that controls which substances can pass from your bloodstream into your brain tissue. The BBB exists to keep toxins, pathogens, and large molecules out while letting essential nutrients (glucose, amino acids, oxygen) through.
GABA is a relatively large, charged molecule. Traditional neuroscience held that oral GABA cannot cross the BBB in significant amounts. This would mean that swallowing a GABA capsule is essentially pointless because the GABA never reaches the brain cells it needs to affect.
However, more recent research has complicated this picture:

The honest answer: we do not definitively know whether oral GABA supplements reach your brain in meaningful amounts. The science is genuinely unsettled. Anyone who tells you it definitely works or definitely does not work is oversimplifying.

Even if GABA does not cross the BBB efficiently, it may still have calming effects through a different route. Your gut has its own nervous system (the enteric nervous system) containing millions of neurons, many of which have GABA receptors.
